PSVIII-B-14 Peroxisome Proliferator-Activated Receptor Gamma Activation Protects Against Deoxynivalenol-Induced Intestinal Barrier Dysfunction in Jejunal Ipec-J2 Cells

Abstract The role of peroxisome proliferator activated receptor gamma (PPARg) in the regulation lipid metabolism, adipocyte differentiation and inflammatory response has been well characterized. Besides adipose tissue, PPARg is also highly expressed intestine. However, functional intestinal function still remains poorly understood. In present study we sought to understand activation on barrier IPEC-J2 cells exposed (2mM, 24hr) mycotoxin, deoxynivalenol (DON). by rosiglitazone troglitazone dose-dependently increased (P < 0.05) protein expression tight junction proteins (TJP), claudin-3 4. On contrary, abundance 4 was decreased antagonist, T0070907. DON exposure TJP, significantly suppressed transcriptional activity. Consistent with therapeutic benefits agonists, pretreatment agonists (40 mM) reversed reduction induced exposure. Immunofluorescence analysis showed that membrane presence claudin-4 ZO-1 troglitazone. Effect included partial restoration transepithelial electrical resistance (TEER) FITC-dextran permeability have disrupted DON. protective effects agonist against abolished confirming importance observed agonists. This shows activity maintenance epithelial homeostasis function, especially during periods metabolic stress Therefore, pharmacological natural bioactive compounds could be effective preventing DON-induced gut dysfunction vivo.
